4. MANUFACTURING PROCESS

PHASE A - AQUEOUS PHASE PREPARATION

Step 4.1: Prepare Water Phase Base

1. Add 35,760.40 g Purified Water to the main jacketed mixing vessel.

2. Start gentle agitation (100 - 150 RPM) using the planetary mixer.

3. Add 2,600.00 g Propylene Glycol to the vessel.

4. Mix for 3 - 5 minutes until uniform.

5. In-Process Check: Verify solution is clear and homogeneous.

Step 4.2: Dissolve Niacinamide and Panthenol

1. With the main vessel agitating at 100 - 150 RPM, slowly add 1,560.00 g Niacinamide powder to the water phase

over 5 - 10 minutes.

2. CRITICAL: Add powder slowly to prevent clumping. Niacinamide is highly water-soluble.

3. Continue mixing for 5 - 10 minutes until Niacinamide is completely dissolved.

4. Slowly add 1,560.00 g Panthenol (Dexpanthenol) to the water phase over 5 - 10 minutes.

5. Continue mixing for 5 - 10 minutes until Panthenol is completely dissolved.

6. In-Process Check: Solution should be clear and colorless with no undissolved particles.

Step 4.3: Disperse Xanthan Gum (Pre-Thickener Dispersion)

1. In a separate container, combine 104.00 g Xanthan Gum with 312.00 g Propylene Glycol (from the weighed

amount) to create a slurry.

2. Mix thoroughly using a spatula until a smooth, lump-free paste is formed (1:3 powder-to-liquid ratio).

3. Add the Xanthan Gum slurry to the main vessel slowly while maintaining agitation at 150 - 200 RPM.

4. Mix for 10 - 15 minutes to ensure complete dispersion.

5. In-Process Check: Verify no "fish eyes" (gel lumps) are present. Solution should be slightly viscous but pourable.

Step 4.4: Disperse Carbomer 980

1. Sift 156.00 g Carbomer 980 powder through a 40-mesh sieve to break up agglomerates.

2. With the main vessel agitating at 200 - 300 RPM, slowly sift the Carbomer powder into the vortex of the liquid

over 5 - 10 minutes.

3. CRITICAL: Add powder slowly to prevent clumping. Do NOT dump all powder at once.

4. After all Carbomer is added, continue mixing for 15 - 20 minutes to allow full hydration.

5. In-Process Check: The mixture should be a thick, translucent gel with no visible dry powder or lumps.

Step 4.5: Heat Phase A

1. Begin heating Phase A to 70 - 75°C using the jacketed vessel heating system.

2. Maintain gentle agitation (100 - 150 RPM) during heating to ensure uniform temperature distribution.

3. Monitor temperature continuously. Heating time: approximately 30 - 45 minutes.

4. CRITICAL SODIUM HYALURONATE ADDITION: Once Phase A reaches 60 - 65°C during heating, slowly sift 52.00

g Sodium Hyaluronate (50-130 kDa) powder into the vessel over 10 - 15 minutes while maintaining agitation at 150

• 200 RPM. Continue heating to 70 - 75°C while mixing for 20 - 30 minutes to ensure complete hydration.

5. Once temperature reaches 70 - 75°C, hold at this temperature for 10 minutes to ensure complete dissolution and

hydration.

6. In-Process Check: Record actual temperature. Verify gel is uniform and translucent. Niacinamide and Panthenol

are stable at 70 - 75°C for the brief processing time required.

• -PHASE B - OIL PHASE PREPARATION

Step 4.6: Melt Oil Phase

1. Add the following to the separate jacketed heating vessel (25 L capacity):

• 3,120.00 g Caprylic/Capric Triglyceride
• 780.00 g Cetyl Alcohol
• 260.00 g Cetearyl Alcohol
• 780.00 g Steareth-2

2. Begin heating Phase B to 70 - 75°C with gentle stirring (100 - 150 RPM).

3. CRITICAL: All waxy solids (Cetyl Alcohol, Cetearyl Alcohol, Steareth-2, Steareth-21, Glyceryl Stearate) MUST be

completely melted. Heating time: approximately 30 - 45 minutes.

4. Once temperature reaches 70 - 75°C, hold at this temperature for 10 minutes to ensure complete melting.

5. In-Process Check: Verify Phase B is a clear, homogeneous liquid with no solid particles or wax crystals visible.

Record actual temperature.

• -EMULSIFICATION

Step 4.7: Combine Phases A and B

1. CRITICAL TEMPERATURE VERIFICATION: Verify BOTH Phase A and Phase B are at 70 - 75°C (within 3 - 5°C of

each other) before combining. Both phases MUST be at the same temperature to prevent shock crystallization of waxy solids (Cetyl Alcohol, Cetearyl Alcohol, Steareth-2, Steareth-21, Glyceryl Stearate).

2. With Phase A in the main vessel agitating at 200 - 300 RPM, slowly add Phase B (hot oil phase at 70 - 75°C) to

Phase A (hot aqueous phase at 70 - 75°C) over 10 - 15 minutes.

3. CRITICAL: Add Phase B in a steady stream, NOT all at once.

4. After all Phase B is added, increase agitation speed to 300 - 400 RPM.

5. Continue mixing for 10 - 15 minutes to form a preliminary emulsion.

Step 4.8: High-Shear Homogenization

1. Engage the high-shear homogenizer at 5,000 - 6,000 RPM.

2. Homogenize for 10 - 15 minutes while batch temperature is maintained at 70 - 75°C.

3. In-Process Check: The emulsion should be smooth, creamy, and uniform with no visible oil droplets or

separation.

4. Reduce homogenizer speed to 3,000 - 4,000 RPM.

Step 4.9: Neutralize Carbomer (In-Situ Thickening)

1. While the batch is still at 65 - 70°C and under homogenization (3,000 - 4,000 RPM), slowly add 156.00 g

Triethanolamine (TEA) to the batch over 5 - 10 minutes.

2. CRITICAL: Add TEA slowly to ensure uniform neutralization and prevent localized over-thickening.

3. Continue homogenizing for 5 - 10 minutes after TEA addition.

4. In-Process Check: The batch should thicken significantly and become opaque white. Viscosity should increase

noticeably.

5. Disengage the high-shear homogenizer.

6. Switch to planetary mixer at 100 - 150 RPM for cooling phase.

• -COOLING PHASE

Step 4.10: Controlled Cooling

1. Begin cooling the batch using the jacketed vessel cooling system.

2. Maintain gentle agitation (100 - 150 RPM) using the planetary mixer during cooling.

3. CRITICAL: Cool slowly to prevent shock crystallization. Target cooling rate: 1 - 2°C per minute.

4. Monitor temperature continuously.

5. Cool to 50 - 55°C. Hold at this temperature for 10 minutes.

6. In-Process Check: Verify emulsion remains stable with no separation or graininess.

Step 4.11: Continue Cooling to 40 - 45°C

1. Continue cooling to 40 - 45°C.

2. Maintain gentle agitation (100 - 150 RPM).

3. PROHIBITION: Do NOT apply high-shear mixing below 50°C (this will destroy the lamellar structure and cause the

cream to collapse).

4. Once temperature reaches 40 - 45°C, proceed immediately to Phase C addition.

• -PHASE C - COOL DOWN ADDITIONS (HEAT-SENSITIVE ACTIVES)

Step 4.12: Prepare Resveratrol Solution

1. In a separate beaker, add 2,600.00 g Propanediol.

2. Add 260.00 g Resveratrol (98% powder) to the Propanediol.

3. CRITICAL OXIDATION PROTECTION: Perform Resveratrol dissolution under nitrogen blanket or in a closed

vessel with minimal headspace. Purge container with nitrogen gas before adding Propanediol and Resveratrol.

Maintain nitrogen atmosphere throughout dissolution process.

4. CRITICAL: Resveratrol requires a minimum 1:10 powder-to-solvent ratio for complete dissolution.

5. Heat gently to 35 - 40°C using a hotplate with magnetic stirrer (DO NOT exceed 40°C to prevent degradation).

6. Stir continuously at 400 - 500 RPM for 20 - 30 minutes until Resveratrol is completely dissolved.

7. In-Process Check: Solution should be clear to pale yellow with no visible crystals or particles.

8. If crystals remain, continue stirring and heating gently until fully dissolved.

9. Cover container with nitrogen blanket or seal immediately after dissolution to minimize oxygen exposure.

Step 4.13: Prepare Bakuchiol & Botanical Extract Blend

1. In a separate container, combine:

• 260.00 g Bakuchiol (liquid oil)
• 85.80 g Bergenia Ligulata Root Extract
• 85.80 g Cipadessa Baccifera Extract

2. Mix thoroughly using a magnetic stirrer at 300 - 400 RPM for 5 - 10 minutes.

3. Pre-warm this blend to 35 - 40°C to prevent thermal shock when added to the main batch.

4. CRITICAL OXIDATION PROTECTION: Cover Bakuchiol blend container with nitrogen blanket or seal immediately

after mixing to minimize oxygen exposure.

5. In-Process Check: Blend should be a homogeneous liquid with no separation.

Step 4.14: Add Phase C to Main Batch

1. Verify main batch temperature is 40 - 45°C.

2. CRITICAL TEMPERATURE MATCHING: Verify Phase C ingredients (Resveratrol-Propanediol solution and

Bakuchiol blend) are at 38 - 42°C (within 3 - 5°C of main batch temperature of 40 - 45°C) before addition. If temperature differential exceeds 5°C, adjust Phase C temperature accordingly.

3. Reduce agitation speed to 50 - 100 RPM (low-shear sweep mixing only).

4. CRITICAL: Transfer Phase C ingredients to main batch quickly (within 2-3 minutes of preparation) to minimize air

exposure time. Consider using nitrogen-purged transfer vessels.

5. Add in the following order:

a. Slowly add the Resveratrol-Propanediol solution (from Step 4.12, pre-warmed to 38 - 42°C) to the main batch over 5 - 10 minutes. b. Mix for 5 minutes. c. Slowly add the Bakuchiol & Botanical Extract blend (from Step 4.13, pre-warmed to 38 - 42°C) to the main batch over 5 - 10 minutes.

6. CRITICAL: Add all Phase C ingredients slowly to prevent thermal shock and ensure uniform distribution.

7. Continue mixing at 50 - 100 RPM for 15 - 20 minutes to ensure complete incorporation.

8. In-Process Check: Cream should be smooth, uniform, and opaque white with no visible streaks or separation.

• -PHASE D - PRESERVATION & FINAL pH ADJUSTMENT

Step 4.15: Add Preservative System

1. Verify batch temperature is 35 - 40°C.

2. In a separate container, pre-mix the preservative system:

• 416.00 g Phenoxyethanol
• 156.00 g Ethylhexylglycerin
• 52.00 g Sodium Phytate

3. Mix thoroughly for 2 - 3 minutes.

4. Slowly add the preservative blend to the main batch over 5 - 10 minutes with continuous mixing at 50 - 100 RPM.

5. Continue mixing for 10 - 15 minutes to ensure uniform distribution.

6. In-Process Check: Cream should remain smooth and uniform.

Step 4.16: pH Adjustment

1. Take a sample of the cream (approximately 10 g) and dilute with 90 g Purified Water (1:10 dilution).

2. Measure pH using a calibrated pH meter.

3. Target pH: 5.8 - 6.2

4. If pH is below 5.8: Add 10% Citric Acid solution dropwise (5 - 10 g at a time) to the main batch with continuous

mixing. Re-check pH after each addition.

5. If pH is above 6.2: This is unlikely given the formulation. If it occurs, add 10% Citric Acid solution dropwise to

lower pH.

6. CRITICAL: Make pH adjustments slowly. Allow 5 minutes of mixing between additions for equilibration.

7. Once target pH is achieved, continue mixing for 10 minutes to ensure uniform pH throughout the batch.

8. In-Process Check: Record final pH. Verify pH is within 5.8 - 6.2 range.

Step 4.17: Final Cooling & De-aeration

1. Continue cooling the batch to 25 - 30°C (room temperature).

2. Maintain gentle agitation (50 - 100 RPM) during final cooling.

3. Once at 25 - 30°C, stop agitation.

4. Optional but Recommended: Apply vacuum de-aeration (if equipment available) at 50 - 100 mbar for 10 - 15

minutes to remove entrapped air bubbles.

5. Alternative: If vacuum de-aeration is not available, allow the batch to rest undisturbed for 12 - 24 hours for static

de-aeration before filling.

6. In-Process Check: Cream should be smooth, uniform, and free of visible air bubbles.
